To date, no studies have assessed the effect of doxycycline on the tetracycline-regulated expression of the multidrug efflux transporter (MATE), C-mycin-dependent tetracycline efflux transporter, and multidrug efflux pump. In the present study, we determined the effect of doxycycline on the expression of the multidrug efflux transporter, C-mycin-dependent tetracycline efflux transporter, and multidrug efflux pump in the rabbit ileum. Doxycycline treatment was used to treat the rat intestine and inhibit MATE-catalyzed cotransformation by covalent binding with the cationic transporters C-mycin-1, C-mycin-2, and MATE-3. The MATE-catalyzed cotransformation in the rat intestine was inhibited by doxycycline, and C-mycin-1, C-mycin-2, and MATE-3 exhibited greater C-mycin-1, C-mycin-2, and MATE-3-mediated cotransformation activity than the MATE-catalyzed cotransformation activity of the other two transporter. Doxycycline had no effect on the expression of multidrug efflux transporter or multidrug efflux pump in the rabbit ileum in vitro. Doxycycline and C-mycin-1 did not affect the expression of the multidrug efflux transporter, C-mycin-2, or MATE-3 in the rat ileum. These results suggest that doxycycline and C-mycin-1 may be used as complementary drugs to inhibit MATE-catalyzed cotransformation by cotransformation. The results of this study suggest that doxycycline may be used as a single agent to inhibit the expression of multidrug efflux transporter and multidrug efflux pump in the rabbit ileum. These results should be considered when determining the efficacy and safety of doxycycline in the treatment of chronic bacterial infections.

Infections are the major cause of morbidity and mortality worldwide [], with infections involving the gastrointestinal tract [–], lung, and skin [–], particularly pneumonia and bronchitis [–], being the most frequently reported pathogens in humans [, ]. The aim of this study was to investigate the effect of doxycycline (Dox) treatment on the expression of the multidrug efflux transporter, C-mycin-dependent tetracycline efflux transporter, and multidrug efflux pump in the rat ileum.
